Initial Report of the Growth Hormone-Binding Protein(GH-BP) in Human Serum using High pressure liquid Chromatography

ABSTRACT
@EN A technique high pressure liquid chromatography (HPLC) gel filtration was used to evaluate GH-binding proteins in human serum, serum (75¥ìl) was incubated with Biotin-hGH (5ng) at 4C for 24h. The minor (Peac ¥°) and major (Peak ¥±) GH-BP was
well
seperated from free Biotin-hGH (Peak ¥²). Elution profile was monitored in optical density multiscaner system. In chromatography, the molecular weight of Peak ¥° and ¥± appeared about 200K and 60K dalton respectively. In control serum, the
specific
binding of biotin-HGH to Peak ¥±-BP was 59.7+/-0.8% of the optical density. Specific binding of Biotin hGH to Peak ¥±-BP was low during the infant period and progressively increased as age. Using gel filtration system, binding of biotin-hGH by
normal
adult serum was dependent on time (maximum equlibrium was reached in 24h at 4C), temperature (4C>22C>37C) and serum concentration. These data indicated that human sera contain a specific binding protein for hGH. The biological significance of
this
binding protein remains to be investigated. We suggest the measurement of GH-BP ¥± activity by HPLC provides a clear seperation. Our nonradioactive optical detection method by multiscaner was also highly resolution (J Kor Soc Endocrinol
